Proto-Time Lord is just the proper name given to “Human plus Time Lord”. It’s the same thing. River is human but genetically engineered into a Proto-Time Lord. The rest of the Proto-Time Lords were cloned from River’s embryonic DNA making them, like River, genetically engineered humans.
